Semiautomatic growth analysis of multicellular tumor spheroids.

2011

Multicellular tumor spheroids (MCTS) are routinely employed as three-dimensional in vitro models to study tumor biology. Cultivation of MCTS in spinner flasks provides better growing conditions, especially with regard to the availability of nutrients and oxygen, when compared with microtiter plates. The main endpoint of drug response experiments is spheroid size. It is common practice to analyze spheroid size manually with a microscope and an ocular micrometer. This requires removal of some spheroids from the flask, which entails major limitations such as loss of MCTS and the risk of contamination. With this new approach, the authors present an efficient and highly reproducible method to an…

Changes of the ratio between myelin thickness and axon diameter in the human developing sural nerve

1978

Axon caliber and myelin sheath thickness of individual nerve fibers were evaluated in the developing human sural nerve using three different methods of measurement: 1. ocular micrometer evaluation of large fibers, 2. photographic enlargements for evaluating large numbers of nerve fibers of all sizes, and 3. electron microscopic enlargements for more precise measurements in selected nerves. The average axonal diameter doubles from 5 months gestation to about 5 years of age. Large fiber group axons increase, during the same period, by a factor of 3--3.5 with a slight decrease thereafter. The myelin thickness increases more slowly, but continuously, between 5 months gestation until the age of …

Cytomorphometric analysis of squames from oral premalignant and malignant lesions

2011

Objective: Oral cytology is becoming increasingly important in the early diagnosis of oral cancer, including epithelial atypia. The aim of this study is to assess the whether the cytoplasmic diameter, nuclear diameter and their nuclear to cytoplasmic ratio are reliable parameters in exfoliative cytology in detecting premalignant and malignant lesions. Study design: Cell and nuclear diameter were measured from the squames of normal buccal mucosal and of oral leukoplakia, oral submucous fibrosis, oral lichen planus which were histologically benign and oral squamous cell carcinoma were measured using an ocular micrometer disc. The nuclear to cytoplasmic ratio in each of these were deduced from…
